UV spectroscopy UV spectroscopy, a widely used analytical technique in pharmaceutical sciences, involves measuring the way a material in solution absorbs visible or ultraviolet light. UV region of the electromagnetic spectrum spans wavelengths from 200nm to 400nm, while the visible region ranges from 400 nm to 700nm. UV spectroscopy is noteworthy for its unique ability to determine the concentrations of multiple components in a mixture simultaneously. This is particularly important for formulations containing combination drugs, which have two or more active pharmaceutical ingredients (APIs). When the absorption spectra of these drugs overlap, as in the case with cefepime and enmetazobactam, simultaneous estimation can become challenging. In such situations, specific techniques such as Vierordt's method and derivative spectrophotometry are employed to overcome the overlap. When molecules absorb UV or visible light, electrons in their outermost shells are promoted from lower energy molecular orbitals to higher energy antibonding orbitals. This transition results in an absorbance signal that can be quantified and correlated with the concentration of the analyte.
